In Hollywood, tijdens de nacht, raakt Rita, een jonge vrouw, geheugenloos na een auto-ongeluk op Mulholland Drive.
Ze ontmoet Betty Elms, een aspirant-actrice die net in Los Angeles is aangekomen. Met haar hulp probeert Rita haar geheugen en haar identiteit terug te krijgen.
